If it says is 100mm in total measurements, that's mean from the middle of cilinder up to the thumbturn lock should be 50mm Explanation: 50+50mm=100mm. Or from the centre of the lock to centre is same measurements. Hope that helps

On the leading edge of the door there will be the screw which retains the cylinder in place, you simply unscrew this and the cylinder barrel should slide out of the door.
