Q: Does this work with avh 2500 nex?

I purchased it for a different head unit that it worked on and then swapped that head unit for an avh-2500nex. I had to carefully transfer some pins from the harness included with the avh-2500nex to this harness, as the plug is physically the same but lacks all of the necessary wires (reverse signal, parking brake sense)

Q: Will this fit 2000 ES 300?

it fit my 2006 Sienna perfectly but I don't know about the 2000 ES 300.

Q: Which wire is the reverse indicator? I'm trying to install a rear camera and I cannot figure out which one is the reverse trigger.

Compare the harness purchased with the original Pioneer harness. The wire will be attached at the same pin location on the harness purchased as it is on the original Pioneer harness. The manual for my head unit shows that it is the violet/white wire on the original Pioneer harness.
