Does anyone know if the 5 is substantially better than the 4.5 as I prefer the smaller size and front panel volume control?

Depends how much accuracy you need- i'd say the 4.5's are great for general listening-- really good - it just comes with a pleasing touch of detail--good for basic monitoring. The E5's are your more serious professional speakers-bigger dynamic range, more accurate mid and bass detail. I've used both and would not use the 4.5's for professional work but they do kick serious butt in terms of depth, clarity and power for such affordable and pleasantly sized boxes. Neither will give you massive sub bass - woofers are only 4.5 inch and 5 inch respectively- the smaller version has a bass response to around 80hz whereas I can get the E5 showing up at 35hz - still the bass they do produce is very clean and you can really tell what's going on with it even in complex pieces of work with multiple bass lines. Again the E5's are better if you need to produce professional grade work.

Does it have an aux input

Yes - two mini jacks on front of active speaker: one for aux-in, the other for headphones. These speakers are good value.

are these compatible with pioneer ddj200?

Hi! Yes, you can connect the Eris E3.5 to the output from the phone, tablet, audio interface etc you are using with the DDJ 200.

Why do both left and right monitors work from the front aux input but when plugged into my laptop from the back rca input only the left monitor works?

Presumably you are using two RCA plugs for left (white) and right (red) - try reversing them - if only the right channel works then your lead is faulty, if only the left channel works the right channel socket on the left speaker is faulty.
