Actually, gripe water is sodium bicarbonate which helps more for upper gas, burps due to indigestion. These are intended for lower gas I the guts.

When given in large doses or for several days in a row, it can. I never had an issues with it with my son, and he got these drops daily for the first few weeks of his life.
