Uner normal conditions how often does the filter need to be replaced?

I have had the 250 for a few months and a slightly different model, 2 Airmega 100 for about a year. These allow you to change the charcoal filter. I vacuum the screen and filters monthly. With these cleanings and open windows with maybe 2 weeks of A/C in summer I would say the filters last a year. No wild fire smoke, a little second hand smoke from another apartment maybe once a night for 15 minutes in the winter in 1 room. To run at an acceptable sound level 24/7 over many days in heavy smoke conditions or summer I recommend getting MORE capacity than you need if you do anticipate wild fire or other  smoke. I'm in a 700 sq ft apt divided into 1 large and one smaller area. I have the 250 and a100 for the larger section, and a 250 for the smaller section in anticipation of wildfire smoke years like 2020 when we had this for a few weeks. In summer I run at least one at level 2 all the time. I wouldn't be able to stand hearing them on setting 3 for more than a quick clear out,  but on setting 2 they are okay should keep the air clean enough. I bought them slowly over a year's time. They do seem to also act as dehumidifiers - which may or may not be helpful depending on conditions where you live and time of year.

Are the Airmega 250 filters the same as the Airmega 400 filters, i.e., interchangeable?

No, they are not interchangeable!  The 250 & 400 filters are different sizes AND the 400 filters are sold in sets of two, because the 400/400S requires two filters (one on each side), whereas the 250/250S requires only one filter.
