Q: Can this be used while pulling the boat?

We do not recommend trailering your boat with the vents in place.

Q: How do you unlock the assembly? I did a test fit but the parts are now stuck together. I've tried pushing and pulling while unscrewing with no luck.

If I understand you correctly, the jam Nut is locked into the base of the outer plate. Remember, the teeth on the base are intended to pinch or penetrate the outer surface of the fabric, locking the Vent in place. So, in case I would get a large pair of pliers. The kind I referring to are called "water pump pliers". Open the jaws wide enough to grab the lock nut that is jammed into place and with your free hand, hold the top of the Vent and turn the nut with the pliers in a counter clockwise direction. This should undo or back off the locked nut. Now, about those pliers, if you don't have a set, your going to have to buy one. Take the Vent with you when you make a purchase. That way you can check the pliers out before you buy,,,or just put the pliers back on the shelf after you've backed the locked Nut off. Simple

Q: What makes the Vent3 unsuitable for trailering, installed V3's still want to trailer

My pontoon boat is 19' long. When I trailer the boat it is never over 50 miles and I generally travel at about 60 MPH. My cover has triple stitching at the vent slots and I have never had any issues.
